Intrathecal delivery of pharmaceuticals to the brain can greatly benefit from accurate computational models of how drugs are dispersed in the cerebrospinal fluid (CSF). In this work, we will showcase a subject-specific framework using imaging data and computational fluid dynamics (CFD) modeling to analyze and optimize different parameters involved in intrathecal drug delivery for central nervous system (CNS) diseases. The method presented in this work will integrate pre-clinical imaging (MRI and PET-CT scans), CFD modeling, and machine learning. Through the proposed framework, we will show simulations of the mixing of two fluids (drug and CSF) and their interaction with tissue structures in a subject-specific model. Moreover, our understanding of the drug pharmacokinetics in the CSF space and the governing parameters of the system within a subject-specific model will be enhanced using these powerful computational and imaging tools.
